The American Spice Trade Association (ASTA) developed the current white paper to assist the spice industry in validating microbial reduction techniques

Validation Steam Sterilization process

Assessing the effectiveness of a sterilisation system is a preventive measure used to control quality and minimise the risk of process failures or safety concerns associated with the equipment. The evaluation process is typically integrated into product control measures throughout manufacturing, helping maximise process performance from raw material intake to finished product production, while meeting the expectations of both regulatory authorities and consumers.

In principle, the evaluation of sterilisation system performance should focus on critical control points (CCPs) that are used to achieve the targeted reduction of Salmonella or to minimise the presence of other specified target microorganisms.

The objective of evaluating the performance of the sterilization system

There are three primary objectives of sterilisation system performance validation:

  • To verify that the sterilisation process is capable of delivering the required sterility assurance level for the product.
  • To demonstrate that the validated sterilisation process can consistently establish sterility across all production lots.
  • To provide additional confidence in product sterility testing results and the effectiveness of the sterilisation system.

Guidelines for Evaluating the Effectiveness of Sterilization Systems According to FDA Standards

The entire sterilisation system performance validation process is conducted in accordance with quality management system requirements and recognised standards, including ISO 9001, ISO/IEC 17020 and ISO/IEC 17025. To ensure objectivity and impartiality throughout the assessment process, the validation is performed by an independent third party, providing confidence to both manufacturers and end users.

Equipment and facilities subject to sterilisation system performance validation may include:

  • Central Sterile Services Departments (CSSD)
  • Autoclaves and sterilisation units
  • Small- and medium-scale sterilisation equipment
  • Thermal processing equipment operating under validated temperature cycles

Sterilisation system performance validation should be conducted at least once per year, or whenever significant changes are made to the equipment, process parameters, or sterilisation protocols.
