The total oxidisable precursor (TOP) assay is a key technique for identifying unknown or hidden PFAS compounds in environmental samples. Through chemical oxidation using persulfate and hydroxide under heat, precursor compounds are transformed into more stable perfluorinated forms like PFOA or PFOS. These are then extracted and quantified via isotope dilution LC/MS/MS.
TOP analysis has shown that in materials like aqueous film forming foams (AFFF), up to 70% of total fluorinated organics may be overlooked without oxidation. Combining TOP assay results with standard PFAS screening provides a more complete and accurate picture of PFAS contamination.
